A 457 does not automatically lead to PR and you need to apply for a PR visa and there are different ones but if it is the 457 that you intend to use as a stepping stone, have a look @ Employer Nomination Scheme (Subclass 121/856)

There are various eligibility requirements such as being on a 457 for two years and your employer also needs to be willing and eligible.
If they are and you already have a number of years experience and qualifications assessed for a SOL occupation, it is possible to do it quicker.

You'll find a link for dependants @ Employer Sponsored Workers (e457) - Online Applications
Your brother and his family will not likely be approved as dependants and he or his wife would have to have eligibility for immigration in their own right.
Edit: More so than " will not likely be approved " you can consider that he and his family will definitely not be approved as a dependant.

Similarly, it is very difficult to have other dependants other than a wife and immediate children proven to be dependent and especially not your sister and with a sister and a brother not approved, that will also mean your mother would not be approved for she would have your brother and sister with her.

If you put other people on an application as dependants and any one of them get rejected, your application will be rejected too.
